Confocal laser scanning microscope with inverted stand

The Leica SP2 laser scanning confocal is built around an inverted (Leica DMIRE2) microscope stand. The inverted stand has a Solent Scientific incubation system with temperature control for live cell imaging and supports both epifluorescence (RGB) and brightfield observation.

The microscope is equipped with a range of objective lenses from 5x to 63x magnification. The SP2 confocal system has 7 laser lines and 3 PMT detectors which are operated via Leica Confocal Software suite. This software includes modules for 3D/4D reconstruction and animation; spectral scanning; analysis and quantification of colocalisation; spectral unmixing and photobleaching (FRAP, FRET).

Detectors

  • 3 fluorescent light PMTs

The booking will be reviewed and then accepted by a member of Bioimaging Hub staff or an alternative time/date proposed.

Please keep to your allotted booking time. You will be charged for the time that you have reserved the instrument for, irrespective of usage. The minimum booking time is 1 hour.

Cancellations should be made at least 24 hours in advance. If you fail to turn up for a booking you will be charged.

To comply with Cardiff University Health and Safety Policy, individuals must receive appropriate training before using the system and be aware of the risks involved in its use.

Users must:

  1. Receive training from an approved member of  staff (indirect training from other users will not be sufficient) - refer to Training and Support section.
  2. Read and understand the latest Risk Assessment for the Bioimaging Unit.
  3. Complete a Registration of Laser Worker form.

All users are expected to observe the following. If they fail to comply they will lose their access rights:

  • All users must be properly trained and registered to use the system before use.
  • All users must read the appropriate risk assessment for the system before use.
  • Food and drink are not permitted within the facility.
  • All waste material (including laboratory gloves) must be properly disposed of via the correct disposal route.
  • All USB data storage devices must be virus checked before plugging them into the system.
  • The system should be correctly powered down at the end of your session and usage correctly logged in the system log sheets.
