Flow Cytometry Facility

The aim of the Flow Facility is to provide affordable access to flow cytometric analysis and cell sorting to resident scientists at Cardiff University. To achieve this, we have 4 benchtop flow cytometers (3 FACSCaliburs, 1 FACSCan) and a high speed MoFlo cell sorter, situated in 3 buildings across the Heath Park site. The benchtop flow cytometers are run by Dr. Eddie Wang, while the cell sorting facility is provided by Dr. Chris Pepper. There are dedicated staff to carry out cell sorting, but benchtop flow cytometry involves informal training from experienced users. Charges are applied on a no profit basis and are designed to recover costs for servicing and consumables.

Cell Sorting

The MoFlo is currently available to measure up to 9 parameters simultaneously using 6 colours, forward scatter, side scatter and time. Flow sorting has numerous applications including the following:

  • Identification and isolation (purification) of transfected cells (GFP/TFP/BFP)
  • Isolation of sub populations of cells with different phenotypes for downstream analysis (e.g. DNA, RNA, protein assays and cell culture)
  • Cloning (single cell per well)

For use on projects including:

  • Chromosome sorting
  • Sub-clone sorting
  • B and T cell subset sorting
  • GFP/YFP/BFP transfected cell sorting
  • Cell cycle fraction sorting (i.e. G0/G1, S, G2/M)
