Third generation multiphoton microscope
This award will fund a 'third generation' multiphoton microscope at Cardiff University.
This equipment is the third-generation evolution of the Coherent Antistokes Raman Scattering (CARS) and four-wave mixing (FWM) imaging capabilities presently available at Cardiff University.
The equipment will enable rapid label-free chemically-specific CARS microscopy and Stimulated Raman Scattering (SRS) of biomolecules (lipids, proteins, DNA) over a wider vibrational frequency range and with reduced photodamage compared to previous generation systems.
The proposed multimodal platform will also enable background-free sensitive imaging of single small metallic nanoparticles (NPs) via resonant FWM detection in the same instrument and will include state-of-the art fluorescence modalities, namely:
- two-photon fluorescence (TPF)
- lifetime imaging (FLIM)
- fluorescence correlation spectroscopy (FCS)
- super-resolution microscopy via Photo-Activated Localisation Microscopy (PALM).
The CARS/FWM imaging capabilities have generated major research impact within Cardiff, the UK and internationally and are run at full capacity through a wide network of collaborations.
The equipment funded through this award is in response to a high user-demand, and will enable this technology to be expanded into a multi-user facility for multi-disciplinary world-class research in physics, chemistry, material sciences and the life sciences.
